Kennedy's 1960 speech as models.
The speech promises to be the most watched of his candidacy to date.
The campaign moved the site of the address from the 20,000-capacity Pepsi Center to Denver's Invesco Field at Mile High, which seats nearly four times that number.
Earlier, former US President Bill Clinton forcefully endorsed Barack Obama's bid for the White House, telling delegates to the Democratic convention that Obama was "ready to lead America and restore American leadership in the world."
The speech by the godfather of the Democratic Party whipped thousands of delegates into an exuberant frenzy.
Where a night before they had hoisted "Hillary" banners, on this night they waved American flags.
The delegates stood on their feet and roared for nearly three minutes when Clinton walked on stage.
The former president basked in their affection, but after several false starts at his speech, commanded: "Sit down!"
Clinton pushed back on attacks - initiated by himself and his wife during the bitter primary campaign, and later taken up by Republican John McCain - that Obama was ill-prepared for the White House, especially on matters of national defence.
"With Joe Biden's experience and wisdom, supporting Barack Obama's proven understanding, insight and good instincts, America will have the national security leadership we need," Clinton said.
Clinton campaigned feverishly for his wife, Hillary Rodham Clinton, in her long-fought primary battle against Obama, and took her loss hard.
He had not spoken out as strongly in support of Obama since he clinched the nomination in June.
But Wednesday, he was unambiguous in passing the torch of Democratic leadership to Obama.
Senator John Kerry, the one-time presidential nominee whose campaign crumbled under attacks on his military service and consistency, told fellow Democrats on Wednesday that he doesn't recognise Republican John McCain, a man he once considered as a running mate.
Kerry, his party's pick in 2004 who failed to defeat incumbent President George W. Bush, said McCain's changed positions on the Republicans' tax cuts, climate change bills and immigration leave him shaken.
"Let me tell you, before he ever debates Barack Obama, John Edwards.
Their ties have since dissolved as they have sparred on the current war in Iraq.
Earlier, Obama was nominated to the Democratic ticket by acclamation.
Former rival Hillary Rodham Clinton interrupted a state-by-state roll call vote at the Democratic National Convention to ask delegates to make Obama's selection unanimous "in the spirit of unity, with the goal of victory."
They agreed, with a roar.
Competing chants of "Obama" and "Yes we can" floated up from the convention floor as Obama's victory was sealed.
Though the convention vote offered no surprises, its historical importance was undeniable.
It capped the longest, closest US primary race in memory as Obama, a political newcomer, defeated Clinton, the former first lady whose victory once seemed all but assured.
It also meant that Obama, the son of a black Kenyan father and a white American mother, is now one victory from becoming president of a nation where, just decades ago, many blacks were denied the vote.
Obama's prospects in the 4 November election are uncertain.
He is in a tight race with Republican John McCain, a veteran senator and former prisoner of war in Vietnam, who has attacked Obama for his lack of experience.
Obama has had to fend off questions about his patriotism and rumours that he is a Muslim.
Republicans hold their convention next week to anoint McCain as their candidate.
